Tuberculosis (TB) is primarily a disease of the lungs caused by the bacterium Mycobacterium tuberculosis (MTB). It is a very old disease. Tuberculosis is a contagious disease. It is a chronic, cyclic disease, mainly affecting the lung and the associated lymph nodes. Like the common cold, it spreads through the air. Only people who are sick with TB in their lungs are infectious. When infectious people cough, sneeze, talk or spit, they propel TB germs into the air. A person needs only to inhale a small number of these to be infected. Overall, one third of the world's population is currently infected with the TB bacillus. Of the people who are infected with TB bacilli, 5-10% becomes sick or infectious at some time during their life.
MTB BactoFind Kit is an in-vitro nucleic acid amplification test for the qualitative detection of Mycobacterium Tuberculosis in human clinical samples. The assay is based on Real time PCR with Taqman probe chemistry. The MTB qPCR Mix contains a primer pair and a dual labeled target detection probe that is used to amplify a genus-specific region of the chromosome and specificity for the M. tuberculosis complex . The MTB target and Internal Control (DNA-IC) are detected in the FAM (Green) and JOE (Yellow) respectively. The Internal control is designed to efficiently control DNA extraction process and inhibition during the PCR amplification.

Product Specifications
I. Sensitivity
These reagents detect 10 copies/reaction. Sensitivity of the assay was determined by serially diluting the positive control of DNA from 100 to 1 copy/PCR. Sensitivity of the assay was determined as 10 copies/reaction. This means that there is 95% probability that 10 Copies/PCR will be detected.
II. Module Specificity
Test result will be positive, only to CMV. The primers and probes were checked for possible homologies to all published sequences (Genbank) by BLAST analysis to avoid any homology with other organisms. In order to check the specificity of this kit different normal/healthy DNA were analyzed. None of these samples led to a positive band in 1.5% (w/v) agarose gel electrophoresis.
III. Reproducibility
Systematic kit production and strict quality control system eliminate batch variation of products. Thus, users can get reproducible data even though they use different batch of product.
IV. Quality Control
Functionality tested in PCR for specificity, sensitivity and reproducibility for specific probe/primer using hematopoietic cell lines, blood positive and negative samples.

• MTB BactoFind Kit is not intended for screening of blood and blood products for the presence of MTB DNA or confirmation of the diagnosis of infection with MTB. This kit has been validated for use with human serum or human plasma collected in EDTA anticoagulant. Test with other sample types may result in inaccurate results. All reagents may exclusively be used for in vitro diagnostics.
• The product is to be used by personnel specially instructed and trained in for the in-vitro diagnostics procedures only.
• Do not subject reagents to 3 freeze-thaw cycles. Store all reagents on ice throughout the experiment.
• It is important to pipette the indicated quantities, and mix well after each reagent addition. Check pipettes regularly.
• Instructions must be followed correctly in order to obtain correct results.
• This test has been validated for use with the reagents provided in the kit. The use of other Reagents or methods, or the use of equipment not fulfilling the specifications, may render equivocal results.
• Detection of mutation depends on the number of fusion transcripts present in the sample, and can be affected by sample collection methods, patient-related factors (e.g. age, symptoms) or for infection stage and sample size.
• Cross contamination between samples and exogenous DNA can only be avoided by following good laboratory practice.
